I tested with a good operating system and hard drive good, persist problem
these are tests that I have done
I have tried another hard drive flex cable, problem persists

j4501pin 1,2,5,7,9,11 present 5V

Any history fault?
Boot with USB macOS and check if HDD appears in Disk Utility.
Discard an APFS issue...

hard drive usb mac os catalina and internal is recognized in disk utilities apperars in afps format, also I tested with internal hard drive hiigh sierra and it starts well without question mark folder

Is the machine working good now, with other internal HDD?

yes with high sierra format mac os plus, but it does not work afps catalina

To make it work with APFS, need to update BIOS.
Take an empty HDD/SSD and do a fresh install of High Sierra, or Mojave.
Then put back the other HDD.
Maybe need a bit of tweak to accept Catalina.
